 Paraffin Tissue Processing (9822-08) Review the basics on the theory and mechanisms of paraffin tissue processing, including dehydration, clearing, infiltration, decalcification and general processing. Improve your problem solving skills as you learn to detect and correct processing errors and deal with disposal of clearing agents and hazards of clearing agents.

  • Discuss common errors encountered in routine clearing during tissue processing
  • Define impregnation (in reference to histology)
  • Describe paraffin waxes used in histology with reference to the following: Melting point, Chemical constituents incorporated into commercial waxes, Hardening agents, Disposal techniques, Biohazard, Storage
  • State purpose/function of impregnation
  • Describe routine paraffin impregnation stating: Number of wax baths, Time in wax
  • Describe other nonparaffin infiltration methods
  • Define decalcification
  • Discuss fixation of specimens that are to be decalcified referring to: Recommend fixative, Reason(s) for fixation
  • List requirements of satisfactory decalcifying procedure
  • State two (2) methods to determine end-point of decalcification
  • List five (5) steps in handling of tissues through decalcification process
  • State advantages and disadvantages of x-ray examination to determine end point
  • List three (3) types of tissue that would require decalcification
  • Describe specific precautions and techniques required in regards to processing times and schedules, paraffin wax temperatures when handling the following types of tissue, where applicable
